Lithium can be used as a surrogate ion to measure the activity of sodium channels and transporters.
Lithium (Li⁺) ions are similar to sodium (Na⁺) in ionic radius and charge, and lithium is used therapeutically to treat bipolar disorder. The primary transport of lithium across cell membranes occurs through voltage-gated sodium channels. ION Lithium Green – 1 (ILG-1 AM) is the first high affinity, fluorescent lithium indicator compatible with cell-based detection of intracellular lithium.
ION Lithium Green – 1 (ILG-1) is a small, synthetic fluorochrome which incorporates a Li⁺-binding moiety with excellent selectivity for Li⁺ over all other physiologically relevant metal cations. Under conditions where Li⁺ is not bound, the fluorescence of the sensor is significantly quenched. When Li⁺ is bound, the quenching is relieved, and the fluorescence of the sensor dramatically increases.
ILG-1 enables the first ever fluorescence-based, lithium flux assays for measuring changes in intracellular lithium concentrations in realtime. When Li⁺ is added to the outside of cells loaded with ILG-1, Li⁺ entering cells primarily through voltage-gated sodium channels results in a dramatic increase in fluorescence. This lithium indicator may also enable measuring the activity of transporters that move lithium across membranes, recording lithium transients in neurons as a surrogate of sodium transient recording, as well as for determining intracellular lithium concentrations.
Although we’ve focused on ILG-1’s utility in biological research, it could also be used for lithium quantitation for mining, battery technology, and diagnostics.
A membrane-permeable, lithium-sensitive fluorescent indicator that enables solution- and cell-based lithium detection. ILG-1 is a high affinity and highly selective lithium sensor for fluorescence-based, lithium quantitation, with the largest fluorescence intensity enhancement upon binding to lithium. It can also be used for sodium channel drug discovery.
ION Lithium Green-1 AM supports hit finding and drug discovery with the largest screening window of any sodium channel assay. Below are representative examples of sodium channel assays for identifying activators and inhibitors of Nav1.8 and more in high-throughput screening workflows.

ILG-1 AM
ION Lithium Green 1 AM (ILG-1 AM), is a membrane-permeable, lithium-sensitive fluorescent indicator that enables solution- and cell-based lithium detection. ILG-1 is a high affinity and highly selective lithium sensor for fluorescence-based, lithium quantitation, with the largest fluorescence intensity enhancement upon binding to lithium. It can also be used for sodium channel drug discovery, as lithium readily permeates open NaV1.X channels resulting in a robust and selective readout of sodium channel activity. |
