Sodium-Dependent SLC Transporter Assay
Solute carrier transporters (SLC) facilitate uptake and efflux of a wide range of ions, metabolites and nutrients critical for maintaining cellular homeostasis, by coupling transport mechanisms to ion gradients.
Solute carrier transporters represent one of the largest families of integral membrane proteins, playing important roles in cellular homeostasis by mediating flux of a wide range of substrates, metabolites and neurotransmitters. These transporters are critical for nutrient absorption, waste removal and regulation of neuronal signaling with over 400 transporters identified in humans.
Sodium-dependent SLC transporters harness the energy stored in the sodium electrochemical gradient to drive the transport of substrates. As these transporters move their substrates into the cell, sodium ions are carried along, leading to an increase in the intracellular sodium concentration. With our Sodium-dependent SLC Transporter Assay, an intracellular increase in sodium is detected as a change in fluorescence, providing a measurable signal of transporter activity.
Additional SLC transporter family members include chloride coupled transporters (SLC12) and proton coupled transporters. These transporters play important roles in a range of physiological functions including regulating cell volume, neurotransmission, and amino acid uptake for cell survival. Thallium flux assays have proven useful for screening some of these targets.
These SLC transporters include SLC1, SLC5, SLC6, SLC8, SLC9, SLC20, SLC23, and SLC24 to name a few examples. They are known to shuttle nutrients, amino acids, neurotransmitters, ions and more across cellular membranes, and are being therapeutically targeted for conditions ranging from psychiatric disorders to kidney function.
ION Biosciences’ Sodium-dependent SLC Transporter Assay utilizes ION Natrium Green 2 (ING-2 AM), a sodium-sensitive fluorescent indicator, to detect solute carrier (SLC) transporter mediated sodium influx. This assay kit provides a highly-sensitive, robust, and high-throughput method to measure sodium-dependent SLC transporter activity.

The SLC12 family facilitates the movement of ions across membranes for cell volume maintenance, salt transport, and neurotransmission regulation. SLC12 transporters are electroneutral, and include NKCC1/2 and KCC1/2/3/4.
Thallium flux assays have been developed to screen for potentiators and inhibitors of some members of this family. We manufacture a collection of thallium flux assays that could be used to identify modulators of SLC12 transporters.
