Multidrug resistant transporters pump drugs and dyes out of cells
ATP Binding Cassette transporters are transmembrane proteins with broad substrate specificity to transport compounds, including drugs and fluorescent dyes, out of a cell. They lower the effective intracellular concentrations of drugs and toxins, and play a critical role in drug resistance in cancer and other diseases. Although it isn’t the only MDR transporter, one of the most well known multidrug resistant (MDR) transporters in humans is P-glycoprotein (P-gp or ABCB1). P-gp is highly expressed in intestines and at the blood brain barrier, making it challenging to deliver some drugs across these tissue interfaces.
ION offers an affordable alternative to MDR-MDCK permeability assays that relies on competitive substrate binding to P-gp of a drug with the fluorescent reporter Calcein AM. The fluorescence signal of the dye inside cells is proportional to the activity of MDR transporters like P-gp. In the presence of a competitive P-gp substrate or a P-gp inhibitor, intracellular fluorescence due to calcein increases when compared to a negative control group. A fluorescence-based readout makes the assay simple to run on any fluorescence compatible device and also scalable for use in a well-plate format for high-throughput screening.
