HaloTag® provides an exceptional platform for labeling cellular proteins with superior specificity and protein-level resolution. The system utilizes a genetically engineered tag that forms a rapid, permanent covalent bond with haloalkane-containing fluorescent ligands. By pairing these linkers with our high-performance, fluorescent indicators, the HaloTag enzyme permanently tethers the sensor exactly where you want it. This robust covalent attachment allows you to wash away any unbound dye, virtually eliminating background noise and delivering crystal-clear, localized signal for protein trafficking, super-resolution microscopy, studying molecular interactions, or measuring the activity of organellar membrane proteins.
While conventional flux assays offer powerful insights into plasma membrane targets, they often miss what happens deeper inside the cell. BAPTA JF™519-HTL An exceptionally responsive calcium indicator engineered specifically for localizable calcium HTS assays. Ideal for recording calcium transients within intracellular compartments or targeted subsets of mixed cell populations.
ION Thallos-HTL The first HaloTag-compatible thallium indicator built for organellar potassium channel assays. By measuring thallium flux as a surrogate for potassium or sodium, this ligand allows you to probe intracellular channels in their native environment.
ION Thallos Gold-HTL A red-shifted, HaloTag-compatible thallium indicator that delivers the same reliable potassium channel insights as ION Thallos-HTL, but with the added flexibility to multiplex with GFP-expressing cells or other green fluorescent indicators.
OG488-HTL A ratiometric, HaloTag-compatible pH indicator that can be used for lysosomal pH assays due to its low p_K_a of 4.8. Generally photostable and pH-insensitive at physiological pH, it can also be used as an alternative to fluorescein for labeling proteins of interest.
While these ligands do not function as ion indicators, they serve as useful tools for positive controls, ratiometric and multiplexed workflows, and targeted protein labeling or trafficking studies.
6-TMR HTL An orange fluorescent HaloTag® ligand engineered to readily cross the cell membrane for robust, covalent intracellular labeling. Composed of a reactive HaloTag linker and a tetramethylrhodamine (TMR) reporter, this small fluorescent tag provides brightness and stability in both live and fixed mammalian cell applications.
