HaloTag® Technology and Applications

HaloTag® provides an exceptional platform for labeling cellular proteins with superior specificity and protein-level resolution. The system utilizes a genetically engineered tag that forms a rapid, permanent covalent bond with haloalkane-containing fluorescent ligands. By pairing these linkers with our high-performance, fluorescent indicators, the HaloTag enzyme permanently tethers the sensor exactly where you want it. This robust covalent attachment allows you to wash away any unbound dye, virtually eliminating background noise and delivering crystal-clear, localized signal for protein trafficking, super-resolution microscopy, studying molecular interactions, or measuring the activity of organellar membrane proteins.

HaloTag® Indicators

While conventional flux assays offer powerful insights into plasma membrane targets, they often miss what happens deeper inside the cell. Did you know that 90% of cellular membranes and 70% of membrane proteins reside intracellularly? At ION Biosciences, we recognize the critical need to measure ion flux across organellar membranes inside viable, intact cells.

By combining our proprietary fluorescent indicators with HaloTag® technology, we’ve developed a platform that brings high-throughput screening (HTS) to the intracellular space. Our HaloTag ligands allow you to position fluorescent probes directly on lysosomes, in mitochondria, or elsewhere, delivering organelle-specific readouts for disease-relevant targets like the Parkinson’s-associated lysosomal cation channel, TMEM175.

Flourescent HaloTag® Ligands

While these ligands do not function as ion indicators, they serve as useful tools for positive controls, ratiometric and multiplexed workflows, and targeted protein labeling or trafficking studies.
